Abstract

In this study, river water and seafood samples were collected monthly from September to November 2005 to determine the occurrence of non-sucrose fermenting Vibrio spp. in the samples, antibiotic resistance and plasmid profiles. 18 water samples and 24 seafood samples were collected monthly during the period of study. The water samples were collected from Sg. Bako, Sg. Kuap and Sg. Melaban. Seafood samples were collected from Gambir market, Kenyalang Park market, Pertanak market and 3n1 miles market. Thiosulfate-Citrate-Bile Salt-Sucrose (TCBS) agar and CHROMagar Vibrio (CV) were used to detect the presence of non-sucrose fermenting Vibrio spp. such as Vibrio parahaemolyticus and Vibrio vulnificus. Confirmation of Vibrio parahaemolyticus isolates were performed via specific PCR with primers targeting the tl gene. Overall, 70 isolates were analyzed for antibiotic resistance and plasmid profiles. Results showed that more than 50% of the V parahaemolyticus isolates were resistant to ampicillin, bacitracin, carbenicillin, cephalothin, methicillin and streptomycin with multiple antibiotic resistant index (MAR) ranging from 0.25 to 0.67. Plasmid DNA with sizes ranging from 1.1 MDa to 91.2 MDa were found in the V parahaemolyticus isolates. All V vulnificus isolates showed resistant towards ampicilin, bacitracin, carbenicillin, cephalothin and methicillin with MAR index between 0.42 and 0.52. Plasmids found in V vulnificus were 3.7 MDa and 4.8 MDa in size. There were no correlation between the antibiotic resistance pattern and the presence ofplasmid.
