Abstract

A research was carried out to establish the clonal and mono culture of Cyanobacteria and to sequence the 165 rRNA gene from selected Cyanobacteria species. Water samples were collected using 20 um mesh plankton net on the subsurface of the ponds. Single cell of selected Cyanobacteria were isolated and grown into clonal cultures. Standard molecular protocols were carried out namely Polymerase Chain Reaction (PCR), purification of PCR products and sequencing. From this study, mono and clonal culture of Anabaena flos aquae and cylindrospermopsis raciborskii has been established. Phylogenetic tree showed based on the 165 rRNA data Anabaena flos aquae and cylindrospermopsis are both monophyletic and sister taxa to each other.
