Abstract
Metroxylon sagu or normally called as sago is an important edible crop in manufacturing starch. Sago plants are mostly found at the rivebanks and swamps throughout Borneo land. DNA extraction analysis was carried out to discover and isolate high quality of total genomic DNA from sago plants. Leaf of sago plants were randomly collected from four population locations at Sungai Kepala, Sungai Pinang, Sungai Stephen Yong and Sungai Sindang Andeh. The modified CTAB method was used to determine amount of DNA that can to be extracted from the young and old samples. Experiment was conducted using large volume of extraction buffer and washing solution. The result from the gel electrophoresis screening showed that the quantity of DNA extracted from twenty four samples were quite low and not consistent. These were due to several factors such phenolic compounds in materials, cross contamination, volume of extraction buffer and sources of material being used. However, CTAB method was applied better in young leaves rather than old leaves. Therefore, further modifications are needed to improve the amount of DNA from each time extraction and to remove unwanted compounds.
