Abstract
The interest of laccase enzyme as a promising biocatalyst in various applications has been grow extensively throughout the years. Various fungi especially from phylum Basidiomycota and white-rot fungi has shown to produce laccase enzyme. Serial dilution was carried out to isolate laccase producing- fungi from leaf litter and undergoes screening for laccase oxidation by using guaiacol and 0.01% chloramphenicol. SSF method was carried out and from utilising leaf litter as substrate to provide more economical and environmentally-friendly. The laccase activity were measured and fungal isolate, Pyrrhoderma lamaense has the highest laccase activity 0.06042 (U/g) with optimum pH of 9. Visualisation of isoforms were taken from native-SDS PAGE for the fungal samples. BLAST analysis was carried out to search match DNA sequence in GenBank database. The data sample scores 100% for Pyrrhoderma lamaense, from phylum Basidiomycota with its purified molecular weight of 750bp.
