Abstract

As human population increases, the demand for land development also increases, resulted in huge areas of forest to be cleared to make way for development and to supply the demand for wood as timber. This can lead to global warming to worsen and depletion of wood resources. Neolamarckia cadamba (Roxb.) Bosser, which has a rapid growth ability, is a tropical tree indigenous to South and Southeast Asia and can assist in countering these problems by means of reforestation. However, a study has successfully discovered the mutant variant of N. cadamba called Kestamas-19, which is induced using colchicine. Since Kestamas-19 is relatively new and not widely known, there are gap in knowledge regarding Kestamas-19. Therefore, in this study, three different concentrations (2.0, 4.0 and 6.0 mg/L) of thidiazuron (TDZ) were used to determine its effect on de novo shoot regeneration of Kestamas-19 explants. After five weeks of culturing, it is found that none of the explants have induced any shoot for any of the TDZ concentrations. This is due to the concentration of TDZ at 6.0 mg/L being too high that inhibits formation of shoots while at 4.0 mg/L and below being too low for induction of shoot. Therefore, longer period of culture is needed in order to see induction of shoot and to fill in the gap in knowledge regarding the optimum concentration of TDZ to induce de novo shoot regeneration of Kestamas-19.
