Abstract
Bioremediation is a biological alternative that uses living organism such as bacteria and fungi to degrade the hazardous compound in the environment. In addition, bioremediation is less costly and more environmental friendly compared to other techniques. There are several problems faced during bioremediation such as low microorganisms' tolerance in toxic conditions and low reusability for next batch uses in bioremediation process. In this study, Thalassospira profundimaris strain M02 was used to degrade the heterocyclic aromatic compound by using cell immobilization technique. Entrapment method was adapted to maintain cell survivability and improve their reusability during biodegradation process. The cell was immobilized by using 2%, 4% and 6% (w/v) calcium alginate and tested with carbazole. Thus, the concentration of sodium alginate and ratio of the cell number; 3g/L, 5g/L and 7g/L to the immobilized media were tested in order to identify the optimization which effected the performance of degradation process. At the end of this study, the optimized conditions for immobilized cells were 4% (w/v) of calcium alginate with 5 g/L of cell and the reusability can up to three cycles. At the end of the experiment, the performance of immobilized cells was 34.69% compared to free cells which is 44.18% that slightly higher in residual carbazole concentration. Thus, the immobilized cells have higher rate of degradation compared to free cells.
