Abstract

A molecular approach of automated ribosomal intergenic spacer analysis (ARISA) was undertaken to characterize the distribution and genetic diversity of the dinoflagellates, Alexandrium species in Malaysian waters which contribute to paralytic shellfish poisoning (PSP). ARISA was used to provide fundamental information on the genetic variability among different populations at different spatial and temporal. This approach was used to identify hot spot of Alexandrium species biodiversity and rapidly detect the species in environmental samples. Samples collection was done on Alexandrium species from Kuching area. The clonal cultures were isolated and established. Scanning electron microscopy (SEM), light microscopy and epifluorescence microscopy were used for the morphological observation of Alexandrium species. Genomic DNA was extracted by using CTAB method. Genomic DNA was used for ribosomal genes amplification and sequencing. The ARISA genus- primers was designed in silico and optimized inculture and environmental samples. Environmental clone libraries were generated from different sampling locations. The ARISA profiles of the environmental samples then be genotyped and analysed by bioinformatic tools.
