Abstract

The effect of inoculum incubation time of Enterococcusfaecium No. 78 on direct lactic acid fermentation was studied. The experiment was carried out to examine the best inoculum incubation time based on lactate production, cell growth, starch and glucose concentration within 24 hours. The batch fermentation processes were conducted using sago starch as a carbon source at pH6.5, temperature of 30°C and agitation of 500 rpm. The experiment was carried out by bioreactor with controlled pH. In the initial stage of the experiment, maximum cell growth was obtained at 9 hours incubation time with 4.2 x log cells/ml. Another experiment was done around the 9 hours incubation time and it was observed that maximum cell growth was obtained at 8 hours incubation time with 5.5 x 108 cells/ml. The lactate production was 13.944 g/L with 9 hours incubation time of inoculum, while 8 hours incubation time of inoculum produced 10.194 g/L lactate. Overall, 9 hours inoculum incubation time was chosen in the direct lactic acid fermentation utilizing sago starch.
