Abstract

The western tarsier, Tarsius bancanus is a Bomean endemic primate, and it is distributed in Sabah, Sarawak, Brunei, Sumatra and Kalimantan. This animal is the smallest primates, and is recognized by having very large eyes. A molecular approach using mitochondrial DNA (mtDNA) Cytochrome Oxidase I (COI) gene is carried out to study the phylogenetic relationship of T. bancanus in Sarawak. The tissue samples were collected from seven locations in Sarawak. Fourteen samples were analyzed using 481 base pairs (bp) of partial COi. Neighbor Joining (NJ), Maximum Parsimony (MP), and Maximum Likelihood (ML) method were used for phylogenetic tree construction. The phylogenetic trees were constructed using Molecular Evolution Genetic Analysis (MEGA) version 6. The intraspecific relationship of T. bancanus was not fully resolved, as supported by low bootstrap value. Meanwhile, the individuals from different locality shows low genetic variation as most of them are grouped together as a sister clade. Overall, the phylogenetic analysis of T. bancanus using partial mtdNA COI was unable to resolve the intraspecific relationship between individuals of different locality.
