Abstract

This study focused on production of industrial important hydrolytic enzymes which are xylanase and pectinase by Aspergillus flavus, Aspergillus niger and Aspergillus versicolor via solid state fermentation using rice husk, palm oil empty fruit bunch (EFB) and sago hampas as substrates. Fermentation parameters such as incubation period (day), incubation temperature (oC), initial moisture content (%), effect of different urea concentration (%), and effect of different mineral salts (%) was investigated to find for favourable incubation condition for xylanase and pectinase production. Result shown that the optimised solid state fermentation for xylanase production at 398 (U/mL) was 6 days of incubation period, 30 oC of incubation temperature, 70% initial moisture content, using Aspergillus flavus with palm oil empty fruit bunch as substrates. Meanwhile the optimised solid state fermentation for pectinase production at 342 (U/mL) was 6 days on incubation period, 30 oC of incubation temperature, 70% initial moisture content using Aspergillus flavus with sago hampas as substrates. Supplementation of urea and mineral salts was found to cause negative effects in xylanase and pectinase enzyme production in terms of enzyme activity.
