Abstract
Mitochondrial DNA of partial cytochrome oxidase (1) gene was used to infer the genetic diversity of Fejen, arya limnocharis among population from different localities in Sarawak. A total 575 base pair (bp) of the COl gene from 10 sequences of Fejen'arya limnocharis were completely aligned by using Mega 7.0. Generally, Fejen'arya limnocharis was revealed the monophyly. The clades produced from the phylogenetic trees showed not resolve because low of diversity that occur within this species and has lower significance of bootsraps values from UNIMAS, Fairy Cave (Bau), Similajau and Miri. They were not clustered together according to geography (western and eastern). This occurred due to different microhabitat between populations. Only one shared haplotype among this population and has supported by Minimum-spanning network. Nucleotide diversity (n) and net nucleotide divergence (Da) values among all population are low which indicate little differentiation among all these populations. Population of Fejen'arya limnocharis showed low population subdivision with relatively high number of migrant per generation which revealed that most of population are panmictic and least migration event occur. This study proven COI gene could not provide sufficient data for population genetic study of Fejenvarya limnocharis. Further study is recommended in future by using different genetic marker to infer overall population genetic structure of this species throughout Sarawak region.
