Abstract

Mungbean (Vigna radiata (Ll Wilezek) was transformed by using Agroboclerium lume(oeiel1s strain LBA 4404. Plasmid pGPTV-SRN4/ALS was cloned into the Agrobacleriul17 lume(aeiens . . Two types of explants were used in this researches which are the leaf cotyledon and cotyledon attached with embryonalax is (CAEA). These explants were co-culti va ted with Agrobaelerium lume(ociens in th e presence of acetosyringone. Binary vector harbouring luei/erase (LUe:') gene (pGPTV), was tested to examine its effec t on transformation efficiency. The cotyledons from treated and germinated seedlings anal ysed via polymerase chain reaction (peR). The res ult obtai ned fro m the comparison of the efficiency of cotyledon leaf (2-3 stages leaves) and cotyledon attached with embryonal axis were inconclusive . For further studies, the transformation efficiency needs 'to be in creased and also improve on the genomic DNA ex trac tion procedure.
