Abstract

Muslim consumers need to consume `Halal' food in order to follow Syariah Law. However, the detection of non halal source in the frozen food products may be occurred due to the adulteration in the food. Halal sausage, halal meatball, non halal sausage and non halal meatball were tested using Polymerase Chain Reaction (PCR) amplification of mitochondrial DNA. Three types of primers were used which were mitochondria DNA of cytochrome b (two types of primers) and mitochondria 12s rRNA (mt-12s rDNA) (one type of primer). The PCR amplification products from cyt b gene and mt- 12s rDNA were unsuccessful to amplify as there were only primer-dimer band formed on the agarose gel. Protein of halal sausage, halal meatball, non halal sausage and non halal meatball were extracted and Bovine Serum Albumin (BSA) standard curve was plotted based on Bradford protein concentration assay. Protein concentrations in all four samples were calculated and Sodium Dodecyl Sulphate Poly Acrylamide Gel Electrophoresis (SDS PAGE) was conducted for protein analysis on all of four samples. There were difference in the clear and prominent bands on all of four samples.
