Abstract

Leonurus japonicus Houtt. is an aromatic herb known locally as Kacangma is popular as folk medicine in Chinese community and has higher potential to be utilized commercially. As to develop new kacangma varieties with improved properties through molecular breeding, study on in vitro induction of haploid plantlet from pollen grain was carried out. The objectives of this study were to establish surface sterilization protocol for pollen grain as explant and to induce direct embryogenesis to produce haploid plantlet. The pollen grain explants were chosen from whole flower bud of day 3 and opened flower of day 4 of flowering stages separately and were tested on different concentration of Clorox and different exposure period. Best surface sterilization protocol were established for 30% Clorox exposed for 20 minutes for the whole bud and 30 % Clorox for 30 minutes for the opened flower with higher percentage of axenic culture yield of 91% and 93% respectively. No protocols were successfully obtained for embryo production through the induction of in vitro haploid plantlet production via direct organogenesis from pollen grain of L japonicus Houtt.
