Abstract

This study was performed to conduct genetic improvement of the oleaginous yeast Lipomyces starkeyi for the enhancement of lipid production. Strain improvement was carried out via random mutagenesis by using ethidium bromide treatment and mutant selection by using cerulenin. Ten mutant isolates demonstrating normal growth in the presence of cerulenin were considered as good candidates for further experiments. The selected mutant strains were designated as L. starkeyi MV-1, L. starkeyi MV-2, L. starkeyi MV-3, L. starkeyi MV-4, L. starkeyi MV-5, L. starkeyi MV-6, L. starkeyi MV-7, L. starkeyi MV-8, L. starkeyi MV-9 and L. starkeyi MV-10. The strains were studied for genetic variability using a M13-PCR fingerprinting approach with L. starkeyi ATCC 12659 as a reference strain. A total of 11 distinctive PCR patterns were obtained which consisted of 7 to 12 bands with the sizes ranging from 280 to 2000 bp. A dendogram was constructed by the unweighted-pair group average (UPGMA) using linkages available in the PyElph software version 1.3.
