Abstract

Leptospu•osis is a zoonotic disease that is commonly detected m subtropical country and humid tropical climate. The disease is caused by Leptospira which is aerobic, spiral and Gram-negative bactenum„ The genus of Leptospira exists in three groups which are saprophytic, pathogenic and intermediate strains. The main aim for this study was to detect and isolate saprophytic, pathogenic and intermediate Leptospira at selected places of Sarawak, Malaysia. A total of 295 samples of rat, soil and water were collected from Gathng National Park and Juara National Training Sewice Centre. The samples were cultured in EMJH medium with adding of 5-fluorouracil fòr three months for rat samples and 30 days for soil and water samples. Extraction of DNA was done using Wizard TM Genomic DNA Purification Kit. Approximately 0.017% (5/295) samples were detected With positive intermediate Leplospira, The confirmations of positive samples were done by ultilizing polymerase chain reaction targetting 16S rRNA gene. The result of the study indicated that the distribution of Leptospira in natural park is higher compared to National Training Servxee Centre.
