Abstract

This study concerned about the screening and cloning of amylase and ceHulase genes from Botryodiplodia theobromae. This fungus is found throughout the tropics and subtropics using numerous kinds of disease but especially rots of fruits and root crops during storage. In this study, B. theobromae used was isolated from mango and cultured on minimal media, minimal media containing starch and minimal media containing carboxymetylcellulose (CMC) as selective media. The existence of these enzymes was indicated by enzyme degradation of the media that produced clear haloes around the culture of fungus. Molecular analysis that have done on this fungus are: genomic DNA extraction, restriction endonuclease digestion. Polymerase Chain Reaction (PCR). cloning and a few confirmation test for clones. In this study. clones of amylase and cellulase and genes were success produced.
