Abstract

A study was carried out to investigate diarrhetic shellfish poisoning (DSP) causing dinoflagellates, Dinophvsis species in Sarawak coastal waters. Regular plankton samples from Santubong and Samariang estuaries were collected using 20µm mesh-size plankton net from August 2009 to February 2010 fortnightly and plankton sample from Sematan was collected once. Micro pipette isolation technique was used to isolate the cells of Dinophvsis species. The number of the Dinophvsis cells remained low throughout the period of study. The live samples were observed under the light microscopy for the morphology characteristic of Dinophvsis, presence of pigments and surface pore arrangement. Two species of Dinophvsis cells were identified in this study, Dinophvsis caudata and Dinophvsis miles. Coloured particles which are believed to be the photosynthetic pigments were found and the arrangement of the pigments were determined by the available space. Well developed left sulcal list (LSL), anterior cingular list (ACL) and posterior cingular list (PCL) were observed in Dinophvsis caudata and Dinophvsis miles. Areoles were also found on the surface of the thecal plates of the Dinophvsis. Toxin analysis was not able to carry out due to the insufficient of Dinophysis cells obtained in this study. Low density of Dinophvsis indicated the low risk of DSP in the areas of study.
