Abstract

Leptospira spp. cause leptospirosis in human with vary signs from asymptomatic, mild to severe and even death. Leptospira can be classified into pathogenic, intermediate and saprophytic strains. Leptospirosis treatment relies on data related to leptospiral susceptibility to different antibiotics. Antimicrobial susceptibility testing (AST) is generally performed in clinical microbiology laboratory to assess the antimicrobial activities of antibiotics to bacteria. However, the recent availability of AST for Leptospira and sensitivity of intermediate and saprophytic strains to antibiotics are still not well developed. This study concerns the antibiotic susceptibility determination in intermediate and saprophytic strains. Ten intermediate and 10 saprophytic strains were assessed against six antibiotics through broth microdilution method. Serial two-fold dilution of antibiotic was added into Ellinghausen-McCullough-Johnson-Harris (EMJH) broth, then inoculated and incubated overnight at 30 °C in dark conditions. OD42o readings of microplate reader were used to determine Minimal Inhibitory Concentration (MIC50) values. Ciprofloxacin, chloramphenicol, and doxycycline had lower MIC5os than penicillin G, amoxicillin, and cefotaxime. Amoxicillin had the highest MIC50 value, with more than 12 800 μg/mL. The results of this study will provide a baseline information on the AST among Leptospira spp. in Malaysia and worldwide.
