Abstract

Assessment of genetic diversity of saplings for Shorea curtisii Dyer ex King was successfully determined by using DAMD markers. 30 DNA samples, each from before and after logging which collected from Compartment 118, Vlu Sedili Forest Reserve, Johor, Malaysia were used in the study. The Shannon's information index and percentage of polymorphic loci were used to quantify the genetic diversity changes of saplings based on 31, 32 and 27 reproducible fragments or DAMD loci generated by primers M13, 33.6 and YNZ-2, respectively. Results from the present study showed that a significant loss ofgenetic diversity in sapling was observed after a single selective logging event under selective management system (SMS). The percentage of reduction for sapling was 18.7% in Shannon's information index and 13.8% in percentage of polymorphic 10cL The results obtained in this study were concordant with the results obtained earlier by using SSR analysis. Therefore, it suggests that primers Le. M13, 33.6 and YNZ-2 are sensitive in monitoring genetic diversity changes or genetic erosion resulting from logging.
