Abstract

Research on the variety preference for major paddy pest in Sarawak, yellow stem borer, remains largely understudied in Malaysia. This study was conducted to determine the most susceptible paddy variety to yellow stem borer by calculation of the egg numbers laid, to document the life cycle of stem borer and to compare the genetic relatedness of yellow stem borer from different regions in Sarawak, Malaysia. The samplings were conducted at Kampung Skuduk Chupak, Kampung Tanjung Tuang and Kampung Pueh from September 2016 until April 2017. For variety preference study, eight varieties were used that are Kanowit, Impun, MR219, Pulut, Gintik, Pulut Hitam, Bario and Hitam (wangi). 12 randomized block design were used and 20 individuals were collected to release into each block. Overall, results showed that MR219 were highly susceptible to the yellow stem borer compared to other varieties tested. For the life cycle study, it can be deduced from the result that a range of 39 to 43 days is needed for an egg to reach 5th instar. However, the individuals were not successfully reached up to pupae and adult stages. Five individuals of yellow stem borer from each region were further used for genetic relatedness study. However, it is suggested from this study that the RAPD primers used were not reliable for yellow stem borer resulted in no PCR product was formed.
