Abstract

Sago palm (Metroxylon sagu) is one of the local and indigenous crops in Sarawak which is grown widely in areas such as Mukah due to its commercial value. Numerous studies have been carried out on sago palm in Sarawak as sago is one of the most important agricultural export commodities of Malaysia. This study was carried out to assess the polymorphism of two types of sago palm which include non-thorny trunking sago palm and non-thorny nontrunking sago palm by using ISSR, SSR and DAMD markers. Genomic DNA of sago palm was successfully extracted by using CTAB method with some modification. The extracted genomic DNA was further amplified using ISSR-PCR, SSR-PCR and DAMD-PCR. PCR amplification using ISSR, SSR and DAMD primers showed the absence of polymorphic band between the two types of sago palm.
