Abstract

Infectious laryngotracheitis (I LT) is an acute respiratory disease aflecting chickens. The disease is caused by infectious laryngotracheitis virus (IL TV). The detection of ILTV by polymerase chain reaction (PCR) has not been reported in Sarawak as no major outbreaks have occurred. This study reports the application of PCR in an attempt to detect the presence of ILTV in village chickens in Kuching Division. The external observation and post-mortem examination had been performed on selected village chickens for finding the symptoms and signs that were related to I LTV. The total DN A had been extracted from 46 serum samples, 46 blood samples and 48 laryngeal tissue samples by henollchloroformfisoamylalcohol (PCI) extraction method. No symptoms of ILTV on village chickens and no signs of IL TV in trachea and laryngeal tissue had been found. The extracted DNA was amplified using ILTU2 and ILTL2 primers via PCR. The PCR analysis shows no live and dormant ILTV in the village chickens in Kuching Division. The positive control that was used in this study is modified attenuated ILTV vaccine. A band pattern from the positive control could be observed after visualization under Ultraviolet (UV) transilluminator. Further similar studies can be performed in other divisions for determining the geographical distribution o f J LTV in village chickens in Sarawak.
