Abstract

Orhtosiphon aristatus (Blume) Miq. is a perennial herbaceous plant that belongs to the family Lamiaceae. There are two varieties found in Malaysia which are the white-corolla variety and purple-corolla variety. These two varieties have similar vegetative characteristics and hard to be differentiated. O. aristatus has very high medicinal properties that made scientists feel interesting to study its breeding system. Improvement of yield and medicinal value of this plant is also crucial to meet demands in market. However, the breeding study is facing problems due to limited flower bud samples used in previous study to reconfirm O. aristatus is a self-pollinated species. Three types of artificial pollination were conducted to study the breeding system of O. aristatus which are selfpollination test, cross-pollination test and apomictic test. From the study conducted, O. aristatus was most likely to be predominantly self-pollinated species based on higher percentage of success rate in seed production from selfpollination test and its flower morphology. Based on the previous studies of pollen viability test, unoptimized media (with or without boric acid) was used to determine the pollen viability of plant from three different flower developmental stages with limited flower tested. Media with different concentration of boric acid were used to determine at which the pollen is most viable by using in vitro germination method through this study. However, no pollen germination found in all boric acid media for each flower developmental stages. Two-dimensional paper chromatography was conducted to obtain flavonoid marker for O. aristatus by using young and mature root samples. In the phytochemical marker analysis, the current study showed polymorphism for OAV-1 and OAV-2 by using mature root samples which might be used as marker in future breeding system.
